Fungal ID
Identification of both yeasts and filamentous fungi begins with DNA extraction from pure isolates using a modified version of the Applied Biosystems PrepMan™ Ultra procedure. The D2 expansion segment of the large subunit rRNA gene is amplified by PCR and the purified PCR product is then sequenced using state-of-the-art fluorescent di-deoxy terminator cycle sequencing chemistry. The extension products are further purified and the sequence is determined on an automated DNA sequencer.
Results using comparative DNA sequences are obtained in anywhere from
7 hours to 14 days, dependent on the turnaround time requested for each
sample. The process normally requires very few cells. Standardized growth
conditions are not required and the technology works equally well with
healthy, damaged, and non-viable cells.
