Sample Preparation
Accugenix has developed and introduced a new, simple method for preparing your samples
which eliminates the need to send samples as traditional cultures. Our new process uses
FTA® Elute Micro Cards to prepare DNA for your samples and makes shipping easier and
less expensive. This method is ideal for international shipping and provides many benefits
for domestic shipping as well. For information on how to submit samples for identification
using FTA® Elute Micro cards, please click
here.
For submitting traditional cultures on plates, slants, etc. please follow the guidelines
below.
Pure cultures are essential for microbial identifications and characterizations.
When submitting a plate with multiple organisms on it, well isolated colonies are required
to avoid the need to subculture for isolation.
For identification by comparative sequencing, Accugenix will extract DNA from original
cultures, and acceptable samples can be cultures growing on a plate or slant, cell pellets
in a plastic tube, or purified DNA. Cultures do not need to be viable to be submitted for
identification.
For strain characterization by automated ribotyping, a live organism less than 72 hours old
is necessary for testing. When submitting a sample for 1 Day Turnaround Time (TAT), it is
essential that a pure culture less than 72 hours old is submitted. For all other TATs, pure
cultures less than 72 hours old are recommended. Any cultures submitted that are more than
72 hours old will require a subculture. TAT for these samples will begin after successful
subculture. Please label each sample with date cultured.
Plates or slants should be sealed with laboratory film. Cultures should be enclosed in a
watertight bag or container (bubble wrap, taped securely, can be used instead).
Liquid cultures are also acceptable, but may add at least one business day to the TAT if the
organism will have to be cultured to ensure purity. Include absorbent material inside the
bag or container (e.g., paper towels or cotton gauze). Please contact Accugenix prior
sending liquid cultures.
Note: ACCUGENIX cannot accept organisms that must be handled in BSL-3 or higher.
Guidelines for International Shipments
Accugenix recommends the use of FTA® Elute Micro Cards to prepare
samples for international shipments. When used with the method developed at
Accugenix, these cards provide a safe and easy way to send DNA for your
samples to us for identification. This method will help to expedite
shipping and comply with U.S. restrictions on shipping known or infectious
agents. Please click here
to find out more about FTA® Elute Cards and
to access information about how to use them.
As an alternative, you may also send DNA prepared with PrepManTM Ultra
Sample Preparation Reagent. Please click here to
access this procedure.
